<p>Every longevity podcast has someone selling an NMN bottle. The bottle costs $80. The protocol calls for two a month. You will hear that it reverses the biological clock, restores mitochondrial function, and adds years to your healthspan. You will not usually hear what Harvard Health, the National Institute on Aging, and the NIH actually say about it.</p>
<p>The pitch is built on real biology. NAD+ is a coenzyme that every cell uses to convert food into energy, and its levels do drop with age. The question is whether swallowing a precursor pill once a day actually changes anything you would feel, see, or measure. Here is what the Tier 1 research shows and what the $300 a month is really buying.</p>
<h3>What NAD+ Is (And Why It Drops):</h3>
<p>NAD+ stands for nicotinamide adenine dinucleotide. Every cell in your body uses it to run mitochondrial energy production, DNA repair, and circadian signaling.</p>
<p>Levels of NAD+ in tissues like skin, brain, and muscle decline meaningfully between roughly age 40 and 60. The exact percentage varies by tissue and study (<a href="https://www.nia.nih.gov/news/nicotinamide-supplement-aging-mice-shows-some-health-improvements-not-increased-longevity" target="_blank" rel="noopener">NIH, 2024</a>). That decline is real. What is contested is whether the decline causes aging or simply reflects it.</p>
<p>If NAD+ levels themselves are the problem, raising them with a supplement should help. If they are downstream of older mitochondria, raising them may do little. Human research has not settled this question.</p>
<h3>Why NMN Got the Hype:</h3>
<p>NMN, or nicotinamide mononucleotide, is one of several NAD+ precursors. The other big one is NR, or nicotinamide riboside. Mouse studies from Harvard, Washington University, and a handful of other labs showed that NMN extended median lifespan in mice and reversed several markers of aging in old animals.</p>
<p>That research generated the podcast surge. By 2024, NMN was a $400 million global market growing about 30 percent a year.</p>
<p>Mouse data is hypothesis-generating, not conclusive. Mice live two to three years. Humans live ten times longer. Drugs that look like reversal agents in rodents routinely fail to show the same effects in human trials, and the longer the disease timeline, the worse the translation tends to be.</p>
<h3>What the Human Trials Actually Show:</h3>
<p>The honest summary so far: NMN supplementation reliably raises blood NAD+ levels in adults. Beyond that, the picture is mixed.</p>
<p>Smaller randomized trials have shown modest improvements in walking endurance, insulin sensitivity, and self-reported sleep at doses around 250 to 600 milligrams daily. Other trials at similar doses found no functional difference versus placebo. None of the human trials to date have run long enough or large enough to test whether NMN extends healthspan or lifespan in people. The trials measure weeks to months, not decades.</p>
<p>Harvard Health summed it up in a 2024 review: the existing human evidence is "preliminary at best," and "no human study has shown that taking NAD precursors slows aging or extends life" (<a href="https://www.health.harvard.edu/staying-healthy/more-evidence-that-aging-might-be-reversible" target="_blank" rel="noopener">Harvard Health, 2024</a>).</p>
<h3>The $300 Question:</h3>
<p>Pharmaceutical-grade NMN at 500 milligrams a day from a reputable brand runs about $80 to $150 a month. Brand-name protocols stacked with resveratrol, TMG, and other "support" compounds push the monthly cost above $300. None of those stacks have been tested as combinations in human trials.</p>
<p>If you are over 50, healthy, sleeping seven hours, and lifting weights twice a week, you may not feel a difference at any dose. If you are under-sleeping, under-exercising, or eating an inflammatory diet, fixing those will affect NAD+ levels more than any pill (<a href="https://www.nia.nih.gov/health/vitamins-and-supplements/dietary-supplements-older-adults" target="_blank" rel="noopener">NIA, 2024</a>).</p>
<p>The NIA's position is that no supplement has been shown to slow aging in people. They recommend treating supplement spend the way you would treat any unproven intervention: small dose, fixed trial window, honest assessment, willingness to stop.</p>
